Methods and suggestions for liquid nitrogen resuscitation samples:
Method 1:
Prepare two sheets of absorbent paper and one clean towel.
1. Spread two layers of clean absorbent paper on the ground or on the desktop.
2. Quickly remove the freezing tube from the liquid nitrogen tank and put it on the absorbent paper (pay attention to the freezing tube, do not stand upright, because the liquid nitrogen in the vertical freezing tube is difficult to volatilize in a short time, and after leveling, the liquid nitrogen will quickly leak out from the gap of the tube).
3. Cover the frozen tube sample with a clean towel (or to prevent the explosion from causing damage), and follow-up tests can be carried out in about 3-5 minutes.
Method 2:
1. According to the strict steps of freezing storage (put the freezing tube at 4, 30 minutes - > - 20, 60 minutes - > - 80, overnight - > liquid nitrogen tank) step by step.
2. After tightening the lid, wrap it around with white tape, which is available in general drugstores. It's the kind of white tape that sticks needles to a bottle. It would be better, but it would still get in. Take out the basket slowly from the liquid nitrogen tank, then take out the freezing tube and put it down at room temperature for more than ten seconds. (Be careful not to put the freezing tube upright.) Melt in 37 degrees water with a cotton gloved hand on the top of the freezing tube.
3. Protective clothing must be worn throughout the operation, including gloves, face masks, etc.
Method 3:
With Nunc freezing tubes, mechanically sealed freezing tubes must be sealed in protective bags. Nunc provides CryoFlex sealed pipes. Thermally sealed polyethylene pipes are specially designed for Nunc freezing pipes. It is very dangerous to freeze the cryopreservation tube in liquid nitrogen without using CryoFlex sealing tube. CryoFlex sealing tube must be used to heat seal the cryopreservation tube in liquid nitrogen.
Warning: Do not use Nunc's freezer in liquid nitrogen unless it is properly sealed in NuncCryoFlex Tubing (Cat. No. 343958). If the cryopreservation tube is not properly used, liquid nitrogen will enter the cryopreservation tube, resulting in pressure rise, explosion or biological leakage safety accidents. The correct use can be referred to NalgeNunc International Cryopreservation Manual.
When the cryopreservation tube is stored in liquid nitrogen instead of using CryoFlex Tubing, the cryopreservation tube should be removed from liquid nitrogen and stored in liquid nitrogen for 24 hours after recovery, which can make the liquid nitrogen evaporate and escape slowly from the cryopreservation tube, thus reducing the pressure in the cryopreservation tube. After 24 hours of storage in liquid nitrogen vapor phase, the frozen storage tube was put into the water bath, covered with a towel, and the frozen storage was transferred after the melting recovery. This method can also be used as pretreatment in method 1 and method 2.
